چکیده
The genetic and antigenic drift associated with the high pathogenicity avian influenza (HPAI) viruses of Goose/Guangdong (Gs/GD) lineage emergence vaccine-resistant field underscores need for a broadly protective H5 A vaccine. Here, we tested experimental vector herpesvirus turkey (vHVT)-H5 vaccines containing either wild-type clade 2.3.4.4A-derived inserts or computationally optimized reactive antigen (COBRA) challenge by homologous genetically divergent HPAI Gs/GD in chickens. Direct assessment protection was confirmed all constructs, which provided clinical against heterologous significantly decreased oropharyngeal shedding titers compared to sham cross reactivity assessed hemagglutinin inhibition (HI) focus reduction assay panel phylogenetically antigenically diverse strains. COBRA-derived elicited antibody responses strains, while wild-type-derived mostly close related clades 2.3.4.4A 2.3.4.4D viruses. In conclusion, HVT vector, widely used replicating vaccine platform poultry, insert provides significant viral challenge. addition, have potential be distinct co-circulating future variants.

Characterization of host responses against a recombinant fowlpox virus-vectored vaccine expressing the hemagglutinin antigen of an avian influenza virus.
There currently are commercial fowlpox virus (FPV)-vectored vaccines for use in chickens, including TROVAC-AIV H5, which expresses the hemagglutinin (HA) antigen of an avian influenza virus and can confer immunity against avian influenza in chickens. Despite the use of recombinant FPV (rFPV) for vaccine delivery, very little is known about the immune responses generated by these viruses in chic...
متن کاملImmunization of Chickens with Newcastle Disease Virus Expressing H5 Hemagglutinin Protects against Highly Pathogenic H5N1 Avian Influenza Viruses
BACKGROUND Highly-pathogenic avian influenza virus (HPAIV) and Newcastle disease virus (NDV) are the two most important poultry viruses in the world. Natural low-virulence NDV strains have been used as vaccines over the past 70 years with proven track records. We have previously developed a reverse genetics system to produce low-virulent NDV vaccine strain LaSota from cloned cDNA. This system a...

Newcastle disease virus (NDV)-expressing avian influenza virus (AIV) hemagglutinin (HA) of subtype H5 was constructed by reverse genetics. A cloned full-length copy of the genome of the lentogenic NDV strain Clone 30 was used for insertion of the ORF encoding the HA of the highly pathogenic AIV isolate A/chicken/Italy/8/98 (H5N2) in the intergenic region between the NDV fusion and hemagglutinin...
متن کاملCan high-pathogenic avian influenza viruses with novel hemagglutinin serotypes other than H5 or H7 emerge?
The family Herpesviridae is a large, diverse family of double-stranded enveloped DNA viruses. Herpes B virus (BV) and Cercopithecine herpesvirus 2 (CeHV-2) are simian herpesvi-ruses. Like herpes simplex virus 1 (HSV-1), they belong to the a-herpesvirus subfamily. interest in BV infections results from the observation that zoonotic infections often result in death of humans, whereas infection of...
متن کاملAnalysis and Professional Designing of COBRA (Computationally Optimized Broadly Reactive Antigen) Vaccine for Bm86 midgut Protein of R. microplus and R. annulatus Ticks
Introduction: The cattle tick Rhipicephalus spp. causes significant economic losses due to diseases in animals and human. Bm86 is a midgut protein and vaccine candidate, which its sequences among the isolates of Ripsephalus spp are geographically separated, variable, and are the main reason for reducing effectiveness, and subsequently, the failure of the recombinant vaccines. Method: In this bi...
